Activation of Inducible Nitric Oxide Synthase by Interferon-¥ã Plus Tumor Necrosis Factor-¥á in Sertoli Cells

Abstract
To study the nitric oxide(NO) synthesis and its signalling mechanism in TM4 Sertoli cells, the cells were treated with recombinant tumor necrosis factor-¥á (rTNF-¥á), recombinant interleukin-1 ¥á (rIL-1¥á), or lipopolysaccharide(LPS), either alone or in combination with recombinant interferon-¥ã (rIFN-¥ã), and NO synthesis was measured by using the Griess method. TM4 Sertoli cells produced a small amount of NO with the treatment of rIFN-¥ã. The effect of rIFN-¥ãwas drastically increased by cotreatment with rTNF-¥ã in a dose-dependent manner, However, combinations of rIL-l¥á or LPS with rIFN-¥ã did not synergies to activate cells. RIFN-¥ã in combination with rTNF-¥á showed marked increase of the expression of iNOS protein. Protein kinase C inhibitors, staurosporin and polymyxin 5, did not inhibit the synthesis of NO induced by rIFN-¥ã plus rTNF-¥á These results suggest that the role of TNF-¥á is to provide TM4 Sertoli cells with the active cofactor for NO synthesis and TNF-¥á-induced signaling for induction of NO synthesis is not dependent on protein kinase C activation.
